Frozen Section

Frozen Section can sometimes be obtained from tissue within minutes. This method of obtaining a session on an emergency basis is called the Frozen Section Technique. This method is used when an early diagnosis of the disease is required, i.e. an emergency report is required to complete the operation to the surgeon during surgery. In this way, sometimes the presence of certain chemicals in the biopsy is checked. which are wasted during normal paraffin wax. So Frozen Section Technique is used. The texture of the section obtained in this method may change because embedding is not done during sample cutting 7 . Since the specimen is not fixed, a special type of instrument called Crystat is used to obtain frozen sections.

Tissue Preparation

A specific part of the biopsy received is excised and with the help of a sharp knife or knife Cut in block form. The thickness of the tissue block should be 5mm. Its sides should be smooth and parallel. As soon as a tissue sample is received that should be processed first, it is best for the surgeon to notify the laboratory before sending the sample so that the laboratory staff can make the necessary preparations. After cutting the received sample or putting the show block into the Test Tube or Biker Boil for 30 to 60 seconds with fixative.

The section is obtained on the slide with great care because the sections are not as rigid as paraffin sections. A plate is usually attached to the Crostat to lift the section. on which the slide is placed. A section is obtained from the tissue through the blade and transferred directly to the slide. The slide is air dried for 30 to 60 seconds. So the action sticks to the slide.

The basic principle of procedure used to stain frozen section is paraffin wax section.

  1. Fix the air-dried section in acetone for 15 – 20 seconds.
  2. Wash the section with water.
  3. Add stain in Hematoxylin for 1-2 minutes.
  4. Wash in water for 5 – 10 Second.
  5. Stain with Eosin
  6. Dip once or twice.
  7. Wash with water.
  8. Dehydrate with alcohol.
  9. Clear from Xylene.
  10. Mount with the help of Canada Bolson.
